To begin, you may speak to Bookie to initiate the dialogue and accept the challenges. You can then put down a bet. Roast Mancomb during th dialogue and you can win the fight this way without having to fist fight. Track the quest to fin the locations of the other challenges.
Afterwards you will need to Defeat Still Waters in a fistfight.
When you face Colusses, he will have a challenge for you. Accept it to face him in a Parry duel and you won't have to fistfight him. You will need to Parry him 3 times. If you refuse the challenge, you will hace to beat him in a fistfight.
